to
http://www.simplyquote.me/quotes/portfolio-item-one ?

Yes that is possible, here follow these steps:

Basically go to Theme Options > Portfolio Settings > Portfolio Page and set custom slug for portfolio item in "Custom portfolio item URL prefix" field.
